5-Vinyl-uridine (VU) is a modified uridine nucleoside bearing a terminal alkene for monitoring RNA transcription. RNA polymerases incorporate it into newly synthesized RNA in place of endogenous uridine. The vinyl group then reacts with labeled tetrazines through IEDDA cycloaddition, enabling rapid, sensitive detection of labeled RNA by fluorescence microscopy or flow cytometry.
