N-azidoacetylglucosamine-tetraacylated (Ac4GlcNAz) is an analogue of N-acetylglucosamine bearing an azide on its N-acyl side chain. Acetylation aids cell membrane permeation; after cellular entry, deprotection allows participation in the hexosamine biosynthetic pathway through the action of GlcNAc kinase.
The resulting modified proteins can be detected by coupling fluorescent tags through Cu(I)-catalyzed azide-alkyne cycloaddition, using the azide functionality as the labeling site.
