sulfo-Cyanine5.5 tetrazine combines a hydrophilic far-red fluorophore with a tetrazine group for click-chemistry labeling. Four sulfo groups contribute hydrophilicity and negative charge, helping reduce nonspecific binding. The reagent is soluble in Water, DMF, DMSO and is suitable for labeling biomolecules in aqueous environments.
The tetrazine group reacts with trans-cyclooctenes, cyclopropenes and some strained cyclooctynes through an inverse electron-demand [4+2]-cycloaddition (IEDDA), producing stable conjugates. This reactivity supports its placement in the tetrazine and cyanine labeling categories.
