Methyltetrazine-PEG12-t-butyl ester is a monodisperse PEG linker combining a methyltetrazine group with a protected carboxyl group. The methyltetrazine supports rapid click reaction with TCO, while acidic conditions can remove the t-butyl protecting group to expose the carboxyl functionality. Its hydrophilic PEG spacer increases water solubility. This combination provides a click-reactive terminus and a separately deprotectable group for subsequent chemical modification.
