PROTAC K-Ras degrader-5 is a cereblon-based PROTAC reported to degrade KRASG12D with a DC50 of <100 nM. It recruits KRASG12D to the cereblon E3 ubiquitin ligase complex, promoting ubiquitination followed by proteasomal degradation.
Reported cellular effects include reduced downstream pERK levels and decreased cancer cell proliferation. In pancreatic cancer spheroids and tumors, it induces caspase 3/7 activity and cPARP, markers of apoptosis.
