APN-PEG4-tetrazine is a heterobifunctional PEG linker pairing a cysteine-selective APN moiety with a tetrazine group. The tetrazine supports rapid, catalyst-free inverse electron-demand Diels-Alder (iEDDA) reactions with strained alkenes such as TCO, norbornene, and cyclopropene, without requiring copper.
The hydrophilic PEG4 spacer improves solubility and reduces steric hindrance during bioconjugation. APN–cysteine conjugates resist hydrolysis in aqueous media, human plasma, and living cells.
