DSPE-N3 is an azide-functionalized phospholipid with a hydrophilic phosphate head and two hydrophobic saturated fatty acid chains connected through glycerol. In water, it can assemble into lipid bilayers for drug encapsulation in liposomes or nanoparticles. Its azide group reacts with cyclooctynes such as DBCO/BCN, or with terminal alkynes in the presence of a Cu catalyst, providing a reactive handle for conjugation.
