ZLC491 is an orally active PROTAC degrader targeting CDK12/CDK13 through cereblon- and proteasome-dependent degradation. It suppresses transcription and expression of long genes, with effects concentrated on a subset of DNA damage response genes. ZLC491 also inhibits proliferation in various triple-negative breast cancer cells, supporting its use in research on this cancer type.
