RBN012811 is a PROTAC-based PARP14 degrader that binds the NAD+ site of PARP14 and forms a ternary complex with cereblon, promoting degradation through the ubiquitin-proteasome pathway. The reported activity is IC50=10 nM, with endogenous PARP14 depletion described in cell lines and primary human macrophages.
Reported cellular effects include reduced IL-10 production and IFN-ß mRNA levels, increased phosphorylated STAT1, and inhibition of interferon-induced ADPr condensate formation. RBN012811 is used to investigate PARP14-related inflammatory signaling, cancer biology, and viral infection, with increased HSV1 replication and reduced VSV replication reported.
