Lurtotecan is a semi-synthetic camptothecin analog listed in the ADC payload category. It stabilizes the topoisomerase I–DNA complex through formation of an enzyme–drug–DNA ternary complex, inhibiting DNA replication and promoting apoptosis. Its described activities also include disruption of RNA synthesis, degradation of topoisomerase I and changes to chromatin structure.
