I-BET 762 is a cell-permeable benzodiazepine compound that binds BET bromodomains. It binds BRD2 (1-473), BRD3 (1-434), and BRD4 (1-477), with Kd = 61.3, 50.5, and 55.2 nM, respectively, by ITC. It also competes with tetra-acetylated H4 peptide for BRD2/3/4 binding and shows little affinity for ATAD2, BAZ2B, CREBBP, PCAF, and SP140.
Reported cellular effects include downregulation of LPS-induced genes and decreased expression of LPS-inducible cytokines and chemokines. This reagent is for laboratory research only; its placement in the PROTAC catalog does not establish degrader activity.
