DBCO-PEG12-Val-Cit-PAB-MMAE is a cleavable ADC linker–payload conjugate combining an azide-reactive DBCO group, a PEG12 segment, a protease-sensitive Val-Cit dipeptide and MMAE through a self-immolative PAB motif. Protease cleavage of Val-Cit initiates PAB fragmentation and MMAE release, while the PEG12 segment increases hydrophilicity.
