Bis-sulfone-PEG4-TCO is a bis-alkylating PEG linker for conjugating the two cysteine thiols produced by reduction of a native disulfide bond, including antibody interchain disulfides. It covalently rebridges these sites through a three carbon bridge while preserving protein structural integrity. The TCO group supports rapid tetrazine click reactions under mild conditions, while the hydrophilic PEG spacer increases aqueous solubility.
TCO can isomerize to the unreactive cis form, limiting retention of click reactivity during storage. Long-term storage is not recommended. The listed storage temperature is -20°C.
