mPEG4-Val-Ala-PAB is a cleavable ADC linker combining a hydrophilic PEG spacer, a Val-Ala dipeptide and a p-aminobenzyl (PAB) group. Its PAB benzylic alcohol provides a site for introducing reactive groups such as PNP for subsequent conjugation to drug payloads. Protease cleavage of the Val-Ala segment enables payload release through an elimination mechanism in the PAB group.
