W146 is a sphingosine-1-phosphate receptor 1 (S1P1) antagonist (Ki = 77 nM) with selectivity over S1P2, S1P3, and S1P5 receptors. In CHO cells, it prevents ligand-induced receptor internalization and inhibits ERK1 and Akt phosphorylation at 10 µM.
Research uses include studies of hematopoietic stem progenitor cell mobilization in combination with AMD3100, as well as endothelial barrier disruption and neutrophil infiltration in mouse models of immune cell-mediated vascular injury.
