16-(Azide-PEG4-ethylcarbamoyl)pentadecanoic-t-butyl ester combines an azide-functionalized PEG segment with an alkane tail bearing a terminal t-butyl ester. The azide reacts with alkyne, BCN or DBCO groups through click chemistry to form a stable triazole linkage, while acidic conditions allow removal of the t-butyl protecting group.
This lipid-PEG linker can spontaneously form micelles in water and may serve in the preparation of more complex lipids for drug-delivery nanoparticles.
