Methyltetrazine-amido-N-bis(PEG4-acid) is a branched PEG derivative bearing a methyltetrazine group and two carboxylic acid groups. It reacts with TCO-containing compounds by inverse-electron demand Diels-Alder cycloaddition, forming a stable covalent bond without copper catalysis or elevated temperatures.
The carboxylic acid groups support coupling to primary amines using activators such as EDC or HATU to form amide bonds. The PEG linker contributes to water solubility; listed solvents are DMSO, Acetonitrile.
