Si5-N14 (solution in ethanol) is an ethanol formulation of a siloxane-incorporated ionizable cationic lipidoid. Si5-N14 has been used to prepare lipid nanoparticles (LNPs) for delivery of mRNA and single-guide RNA (sgRNA) in vitro and in vivo.
In immortalized human lung microvascular endothelial cells (iMVECs), Si5-N14-containing LNPs encapsulating a luciferase mRNA reporter showed greater cellular uptake through micropinocytosis and lipid raft-mediated endocytosis, together with greater endosomal escape, than LNPs containing non-siloxane-incorporated lipids.
