MS154 is a cereblon-recruiting PROTAC degrader that links a cereblon-binding moiety to gefitinib and targets mutant epidermal growth factor receptor (EGFR). In mutant EGFR-bearing lung cancer cells, it reduces EGFR protein levels, suppresses downstream signaling and inhibits proliferation. Reported DC50 values are 11 and 25 nM in HCC-827 and H3255 cells, respectively, with Dmax > 95% at 50 nM. These effects were not observed in the tested WT-EGFR-bearing ovarian and lung cancer cell lines. Bioavailability has been reported in mice following ip administration.
