Mal-amido-PEG8-val-gly-PAB-PNP is a cleavable ADC linker with a terminal maleimide, a PEG chain, a Val-Gly dipeptide, a PAB group, and a PNP group. The maleimide forms covalent bonds with free thiols on protein cysteine residues, while the Val-Gly dipeptide is cleaved by intracellular cathepsin enzymes to support payload release.
