N-(TCO)-N-bis(PEG4-acid) is a branched click chemistry reagent bearing a terminal TCO group and two terminal carboxylic acids. The TCO moiety undergoes rapid, selective inverse-electron demand Diels-Alder cycloaddition with tetrazine-containing molecules to form stable covalent bonds. The terminal carboxylic acids react with primary amino groups in the presence of activators including EDC and HATU.
