NH2-PEG6-CH2CH2COOtBu, also known as Amino-PEG6-t-butyl ester, combines an amino group with a tert-butyl-protected carboxyl group through a hydrophilic PEG spacer. The spacer supports aqueous solubility, while the amino group provides a site for reaction with carboxylic acids, activated NHS esters, and carbonyl compounds such as ketones and aldehydes. Acidic conditions can remove the tert-butyl protection to expose the carboxyl group for further functionalization.
This reagent is used in PROTAC synthesis and as a non-cleavable 6-unit PEG linker in antibody-drug conjugate research.
