CG428 is a CRBN-dependent PROTAC degrader targeting tropomyosin receptor kinase (TRK), with reported anti-tumor activity. In KM12 colorectal carcinoma cells, it reduces levels of the TPM3-TRKA fusion protein (DC50 = 0.36 nM), supporting its use in colorectal carcinoma research.
CG428 binds TRKA more strongly than TRKB and TRKC (Kd = 1 nM, 28 nM and 4.2 nM, respectively). It is cataloged within the TRK receptor degrader group.
