14-(Fmoc-amino)-tetradecanoic acid is an aliphatic linker used in PROTAC synthesis, with an alkane chain bearing an Fmoc-protected amine and a terminal carboxylic acid. Basic conditions remove the Fmoc protecting group to expose a free amine for subsequent conjugation. The carboxylic acid provides a separate coupling site, reacting with primary amines in the presence of activators such as EDC or HATU to form an amide bond.
