KI696 inhibits the interaction between Keap1 and Nrf2 by binding to the Keap1 Kelch domain. In isolated human bronchial epithelial cells, it increases Nrf2 accumulation in a concentration-dependent manner. It also increases expression of the Nrf2 target genes NQO1, GCLM, HMOX1, and TXNRD1 in bronchial epithelial cells isolated from patients with chronic obstructive pulmonary disease. The compound is cataloged among KEAP1/NRF2 inhibitors.
