Azido-PEG4-Val-Cit-PAB-OH is a cleavable ADC linker combining an azide handle, a hydrophilic PEG spacer, a Val-Cit dipeptide and a PAB group. The azide supports click conjugation with terminal alkynes or strained cyclooctynes such as DBCO or BCN. The PAB benzylic alcohol provides a site for activation and subsequent payload conjugation. Intracellular protease cleavage of Val-Cit initiates payload release through an elimination mechanism involving the PAB structure.
