MC-Val-Ala-NHS ester is a cleavable ADC linker combining a Val-Ala segment with maleimide and NHS ester functional groups. The Val-Ala segment is susceptible to cleavage by Cathepsin B, while the maleimide reacts with thiol groups. The NHS ester forms covalent bonds with amines under neutral or slightly basic conditions, including lysine side chains and amines on aminosilane-coated surfaces.
