TCO-PEG4-Val-Cit is a cleavable ADC linker combining a TCO click handle, a hydrophilic PEG spacer, and a Val-Cit dipeptide. The TCO group reacts with tetrazine groups, while the Val-Cit segment is susceptible to cleavage by cytoplasmic peptidases for drug-release applications.
