t-Boc-N-amido-PEG4-Val-Cit is a protease-cleavable ADC linker combining a Boc-protected amine, a hydrophilic PEG spacer and a Val-Cit dipeptide. Cellular proteases can cleave the Val-Cit unit, while the free carboxylic acid provides a site for amide formation with amines. Acidic removal of the Boc group exposes a primary amine for further coupling reactions or reductive amination.
