Fmoc-PEG3-Val-Cit is a cleavable ADC linker combining an Fmoc-protected amine, a hydrophilic PEG spacer and a Val-Cit group. Basic conditions remove the Fmoc protection to expose an amine for subsequent conjugation. The PEG spacer increases aqueous solubility, while the Val-Cit group is susceptible to lysosomal proteolytic enzymes and is reported to remain highly stable in human plasma. Its listed purity is 98%.
