NH2-PEG3-Val-Cit-PAB-OH is a cleavable ADC linker combining a primary amine, a hydrophilic PEG spacer, a Val-Cit dipeptide and a PAB group bearing a benzylic alcohol. The amine supports coupling with carboxylic acids or reductive amination with aldehydes and ketones, while the benzylic alcohol provides a site for activation and payload conjugation.
Cellular protease cleavage of the Val-Cit segment enables payload release through an elimination mechanism involving the PAB structure.
