t-butyl ester-PEG5-CH2COOH is a PEG acid linker with a free terminal carboxylic acid and a second carboxyl group protected as a t-butyl ester. Activation with EDC or HATU enables the free acid to couple with primary amines, forming an amide bond. Acidic conditions can remove the t-butyl protecting group.
