Fluorescein-PEG8-azide is an azide-functionalized xanthene dye with excitation/emission maxima of 494/517 nm. It reacts with terminal alkynes through copper-catalyzed click chemistry (CuAAC) and with strained cyclooctynes through copper-free click chemistry. The latter reaction forms a stable triazole without a copper catalyst or elevated temperatures.
The hydrophilic PEG spacer increases aqueous solubility and reduces steric hindrance during binding. This fluorescein labeling reagent can also be used as a quantum yield standard. Its listed purity is 96%, with storage at -20°C.
