PFI 4 is an SGC chemical probe targeting the bromodomain of the scaffolding protein BRPF1B. Reported binding values are IC50 = 172 nM and Kd =13 nM, with the latter determined by ITC. In a triple BRD cell construct, it reduces FRAP recovery time and shows cellular potency of IC50 250nM, with no effect on BRPF1A. Reported selectivity for BRPF1 is >100-fold over a bromodomain panel including BRPF2 (BRD1), BRPF3 and BRD4.
